Изучение антиоксидантных, гепатопротекторных и противовоспалительных свойств комплекса фенолгликозидов коры осины (англ. aspen bark phenol glycoside complex, ABPG) в моделях in vitro.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Комплекс ABPG выделяли экстракционным методом, компонентный состав определяли методом высокоэффективной жидкостной хроматографии. Антирадикальную активность оценивали в тесте с 2,2-дифенил-1-пикрилгидразилом (ДФПГ), железосвязывающую способность – колориметрическим методом с феррозином. Клеточные эффекты изучали на культуре клеток линии HepG2 в моделях липотоксичности (олеиновая и пальмитиновая кислоты), парацетамол-индуцированного повреждения и воспалительной стимуляции липополисахаридом (LPS) и (или) интерфероном (IFN) γ. Конечными точками служили уровень внутриклеточных активных форм кислорода (АФК), содержание восстановленного глутатиона (GSH) и нейтральных липидов или жизнеспособность клеток.</p></sec><sec><title>Результаты</title><p>Результаты. ABPG содержит более 70 индивидуальных соединений; доминирующими компонентами (по доле площади пиков) являются дигидроксибензойная кислота (11,5%), метоксибензойная кислота (8,5%), лютеолин (7,7%), салицин (5,4%) и гидроксибензойная кислота (1,88%). Комплекс проявлял выраженную антирадикальную активность (IC50 = 87,6 мкг/мл в тесте с ДФПГ) и связывал ионы Fe2+ в количестве, эквивалентном 14,07 мг этилендиаминтетрауксусной кислоты на 1 г образца. В клетках HepG2 ABPG снижал уровень АФК в условиях липотоксичности на 47,6% и препятствовал накоплению внутриклеточных липидов на 40,4%; при парацетамол-индуцированном повреждении повышал жизнеспособность клеток до 77,9%; в условиях стимуляции LPS/IFN-γ снижал продукцию АФК на 19,2%, одновременно повышая уровень GSH на 42,9%.</p></sec><sec><title>Заключение</title><p>Заключение. Комплекс фенолгликозидов коры осины модулирует редокс-состояние и стресс-ответы гепатоцитов, обеспечивая цитопротекцию через редокс-зависимые механизмы. Эти эффекты лежат в основе его антиоксидантного, гепатопротекторного и противовоспалительного действия in vitro.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>The aim</title><p>The aim. To study the antioxidant, hepatoprotective, and anti-inflammatory properties of an aspen bark (Populus tremula L.) phenol glycoside complex (ABPG) in vitro.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. ABPG was obtained by solvent extraction and characterized by high-performance liquid chromatography (HPLC). Radical scavenging activity was assessed by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, and iron chelating capacity was evaluated by the colorimetric ferrozine-based assay. Cellular effects were studied in HepG2 cells in models of lipotoxicity (oleic and palmitic acids), paracetamol-induced injury, and inflammatory stimulation with lipopolysaccharide (LPS) and interferon γ (IFNγ). Intracellular reactive oxygen species (ROS), reduced glutathione (GSH), neutral lipid content, or cell viability served as endpoints.</p></sec><sec><title>Results</title><p>Results. ABPG comprised more than 70 individual constituents; its dominant components (as a share of the total HPLC peak area) were dihydroxybenzoic acid (11.5%), methoxybenzoic acid (8.5%), luteolin (7.7%), salicin (5.4%), and hydroxybenzoic acid (1.88%). The complex showed pronounced radical scavenging activity (IC50 = 87.6 µg/ml in the DPPH assay) and chelated Fe2+ ions in the amount equivalent to 14.07 mg ethylenediaminetetraacetic acid per gram of the sample. In HepG2 cells, ABPG lowered ROS levels under lipotoxic conditions by 47.6% and prevented intracellular lipid accumulation by 40.4%. In paracetamol-induced injury, it restored cell viability to 77.9%; and under LPS/IFNγ stimulation, it decreased ROS production by 19.2% while raising GSH levels by 42.9%.</p></sec><sec><title>Conclusion</title><p>Conclusion. The aspen bark phenol glycoside complex modulates the redox state and stress responses of hepatocytes, conferring cytoprotection through redox-dependent mechanisms.
