Оценка in vitro механизмов гиполипидемического эффекта сесквитерпенового γ-лактона ахиллина на культуре клеток гепатомы крыс.</p></sec><sec><title>Материал и методы</title><p>Материал и методы. Изучено влияние сесквитерпенового γ-лактона ахиллина и гемфиброзила (препарат сравнения) на жизнеспособность, содержание липидов и экспрессию ключевых генов обмена липидов в клеточной культуре гепатомы крыс (HTC). Содержание липидов оценивали флуоресцентным методом с витальным красителем NileRed, жизнеспособность клеток оценивали с помощью МТТ-теста.</p></sec><sec><title>Результаты</title><p>Результаты. Культивирование клеток гепатомы в течение 48 ч сахиллином в концентрации 0,25–1,0 мМ и гемфиброзилом 0,25–0,5 мМ не приводило к изменению жизнеспособности клеток по сравнению с контролем. В этих же концентрациях исследуемые вещества уменьшали содержание липидов в клетках, оцененных флуоресцентным методом с витальным красителем NileRed. Для исследования механизма гиполипидемического действия ахиллина определяли экспрессию ключевых генов метаболизма липидов в клеточной культуре линии НТС. Возможный механизм гиполипидемического действия ахиллина может быть обусловлен увеличением транспорта и окисления длинноцепочечных жирных кислот в митохондриях, о чем свидетельствует повышение экспрессии гена карнитин-пальмитоилтрансферазы2 Cpt2. Снижение уровня холестерола может быть связано с повышенным синтезом желчных кислот из холестерола вследствие повышенной экспрессии гена 7-альфа-гидроксилазы Cyp7a1.</p></sec><sec><title>Заключение</title><p>Заключение. В культуре клеток НТС сесквитерпеновый γ-лактон ахиллин снижает накопление липидов в клетках, о чем свидетельствует уменьшение флуоресценции NileRed, повышение экспрессии генов Cpt2 и Cyp7a1.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Objective</title><p>Objective. Evaluation in vitro of the mechanisms of the hypolipidemic effect of sesquiterpene γ-lactone achilline in the hepatoma tissue culture (HTC).</p></sec><sec><title>Materials and methods</title><p>Materials and methods.The influence of sesquiterpene γ-lactone achilline and gemfibrozil (comparison drug) on the viability, lipid content and expression of key genes of lipid metabolism in the hepatoma tissue culture. The lipid content was assessed by fluorescent method with the vital dye Nile Red, the cell viability was assessed using MTT assay.</p></sec><sec><title>Results</title><p>Results. Cultivation of of cell cultures of rat’s hepatoma cell line HTC for 48 h with achilline in a concentration of from 0.25 to 1.0 mm and gemfibrozil from 0,25 to 0,5 mm did not change cell viability compared to control. In these same concentrations of the test substance reduced the lipid content in the cells, assessed by fluorescent method with the vital dye Nile Red. To study the mechanism of hypolipidemicaction of achillinedetermined the expression of key genes of lipid metabolism in cell culture lines HTC. The possible mechanism of hypolipidemic action of achilline can be attributed to the increased transport and oxidation of long-chain fatty acids in mitochondria, as evidenced by the increase in the gene expression of carnitine-palmitoyltransferase 2 (Cpt2). The decrease in cholesterol level may be due to increased synthesis of bile acids from cholesterol, due to increased gene expression of 7-alphahydroxylase (Cyp7a1).</p></sec><sec><title> </title><p> </p></sec><sec><title>Conclusion</title><p>Conclusion.
